Pronuclear or cytoplasm injection of FVB/NJ zygotes

$4,545 per construct

Over two-three days at least 250 FVB/NJ zygotes will be injected and implanted in at least 10 pseudopregnant recipients. Charges will be made once injections are completed. No guarantee is made about the number of live born mice or the number of positive transgenic founders that will be identified. Clean transgene preparations will normally produce at least 20 offspring. We require an EtBr-stained gel of 100 nanograms of the purified transgene fragment run along side 10 microliters of the ladder (NEB catalog #) to assess the intactness, purity and concentration of the fragment to be injected.

Pronuclear or cytoplasm injection of C57BL/6J zygotes

$5,050 per construct

Over two-three days at least 250 C57BL/6J zygotes will be injected and implanted in at least 10 pseudopregnant recipients. Charges will be made once injections are completed. No guarantee is made about the number of live born mice or the number of positive transgenic founders that will be identified. Clean transgene preparations will normally produce at least 20 offspring. We require a photo of an EtBr-stained gel on which 1-3 ul of the purified construct is run with a ladder with known band concentrations (expect low concentrations ~ 0.5 – 1.0 ng/ml). This allows us to assess the intactness, purity and concentration of the fragment to be injected.

ES cell blastocyst injection

Two days of blastocyst injection are performed for your clone of genetically-modified ES cells. We inject at least 75 blastocysts and implant into at least 5 pseudopregnant recipients. All offspring are transferred to investigator after weaning. Charges will be made once injections are completed. No guarantee is made about the number of live born mice or the quality of chimerism that will be obtained. High quality ES cell lines (e.g. v6.5) normally generate at least 15 offspring.

SPF rederivation

$505/day plus animal purchase and per diems

The MGL has two techniques that may be employed for the rederivation of your mouse lines into SPF status. The techniques include embryo transfer and In Vitro Fertilization (IVF). The MGL requests that you contact us directly to determine the appropriate methods for the mouse lines that you would like rederived. The investigator will be responsible for the direct cost of animals used during these procedures and for the per diems of those animals that are generated during the rederivation process.

Sperm Cryopreservation

$505/1-3 lines

We ask for two proven fertile males, age between 8 - 24 weeks, per mutant line.  The age limitation is somewhat flexible. We combine the sperm from the two males and cryopreserve the sperm in cryoprotectant media, aliquot them into 16 vials (100 ul per vial) and store them in MGL liquid nitrogen tank. Please email Yun You (yyou@umn.edu) directly if you have any question.  In the Request for Service Form, we ask current location of your mice so we can determine if your mice are parvo negative, or suspect, or positive, so that MGL can prepare the cryopreservation procedure accordingly.

In vitro fertilization (IVF)

$1655 plus per diems

This is to recover mouse lines from cryopreserved or fresh sperm. The technical aspect of IVF requires a minimum of two days of service.

Specialized services

$505/day plus mouse purchase and per diems

The MGL is able to adapt basic mouse genetic protocols and embryo manipulation techniques to achieve the specific aims of your projects. The MGL requests that you contact us directly to determine the specific requirements of your project. The investigator will be responsible for the direct cost of animals used during these procedures and for the per diems of those animals that are generated during these specialized services. Examples of Special Service include, but not limited to, transgenic rescue (DNA or CRISPR injection) of PI's mutant zygotes (or specific strain) when mice are housed in PI's animal rooms.  Isolating 2-cell to blastocyst stage mouse embryos, culture or consulting of mouse ES cells or gene targeting mouse ES cells. Ovary transplantation.
